Abstract

ÀÌ ¿¬±¸´Â PGE_(2)»ýÇÕ¼º ¾ïÁ¦Á¦ÀÎ Àεµ¸ÞŸ½ÅÀÇ Åõ¿© ½Ã ¼ºÀå±â °³ÀÇ ÇϾǵΠ¿¬°ñ¿¡ ³ªÅ¸³ª´Â cyclooxygenase-2(COX-2)¿Í insulin-like growth factor ¥°(IGF-¥°)ÀÇ ¹ßÇö°ú ºÐÆ÷¸¦ °üÂûÇÏ¿© Àεµ¸ÞŸ½ÅÀÌ ÇϾǵΠ¿¬°ñ ¼ºÀå¿¡ ¹ÌÄ¡´Â ¿µÇâÀ» ±Ô¸íÇϱâ À§ÇÏ¿© ½ÃÇàÇÏ¿´´Ù. »ýÈÄ 12-13ÁÖµÈ Àâ°ß 8¸¶¸®¸¦ 4±ºÀ¸·Î ±¸ºÐÇÏ¿´´Ù. Åë»óÀû º¹¿ë·®ÀÎ Àεµ¸ÞŸ½Å 2mg/Kg/dayÀ» °¢°¢ 7ÀÏ°ú 14ÀÏ°£ Åõ¿©ÇÑ ±º, °ú·®ÀÎ 8mg/Kg/dayÀ» 14ÀÏ°£ Åõ¿©ÇÑ ±º°ú ¹«Ã³Ä¡±ºÀÎ ´ëÁ¶±ºÀ¸·Î ±¸ºÐÇÏ¿´À¸¸ç, ÇϾǵθ¦ ¿¬±¸´ë»óÀ¸·Î ÇÏ¿´´Ù. ¿¬±¸´ë»ó ÇϾǵδ 5§­µÎ²²ÀÇ ÀýÆíÀ» ¸¸µé¾î H-E Áß¿°»ö COX-2 ¸é¿ª¿°»ö, IGF-¥° ¸é¿ª¿°»öÀ» ½ÃÇàÇÏ¿© ±¤ÇÐ Çö¹Ì°æÀ¸·Î °Ë°æÇÏ¿´À¸¸ç, tartrate resistant acid phosphatase (TRAP) ¿°»ö ÈÄ ÆÄ¿¬°ñ¼¼Æ÷ÀÇ ¼ö¸¦ ÃøÁ¤ÇÏ¿© ´ÙÀ½°ú °°Àº °á°ú¸¦ ¾ò¾ù´Ù. Àεµ¸ÞŸ½ÅÀº ÇϾǵΠ¿¬°ñÀÇ Áõ½Ä´ë¿¡¼­ COX-2¿Í IGF-¥°ÀÇ ¹ßÇö°ú ºÐÆ÷¸¦ ¾ïÁ¦½ÃÄ×À¸¸ç, Àεµ¸ÞŸ½ÅÀÇ Åõ¿©±â°£¿¡ ºñ·ÊÇؼ­ ÆÄ¿¬°ñ¼¼Æ÷ ¼ö´Â À¯ÀǼº ÀÖ°Ô °¨¼ÒÇÏ¿´´Ù(p<0.01).IGF-¥°ÀÇ ¹ßÇö°ú ºÐÆ÷´Â Àεµ¸ÞŸ½ÅÀÇ Åõ¿© ¾ç°ú ±â°£¿¡ ºñ·ÊÇÏ¿© ¾ïÁ¦µÇ¾ú´Ù. ÀÌ»óÀÇ °á°ú¿¡ ÀÇÇϸé Àεµ¸ÞŸ½ÅÀÇ Åõ¿©´Â ÇϾǵΠ¿¬°ñ¿¡¼­ COX-2¿Í IGF-¥°ÀÇ ¹ßÇö°ú ºÐÆ÷¸¦ ¾ïÁ¦ÇÏ°í ÆÄ¿¬°ñ¼¼Æ÷ÀÇ ¼ö¸¦ °¨¼Ò½ÃÄÑ ÇϾǵΠ¼ºÀåÀ» ¾ïÁ¦ÇÒ °¡´É¼ºÀÌ ÀÖÀ½À» ½Ã»çÇÑ´Ù.

This study aimed to investigate the effects of indomethacin on distribution and expression of COX-2 and IGF-¥° in the mandibular condyle of growing dogs and to examine the number of chondroclasts around the mineralization zone. Indomethacin inhibits prostaglandin E_(2)production in the tissue by inhibiting synthesis of cyclooxygenase 2. Prostaglandin E_(2) stimulates insulin-like growth factor synthesis. Insulin-like growth factor stimulates growth of mandibular condyiar cartilage. Eight mongrel dogs. aged 13-14weeks. were divided into 4 groups. Group 1 and group 2 were administered indomethacin 2 mg/Kg/day orally two times a day for 7 days and 14 days respectively. Group 3were administered indomethacin 8 mg/Kg/day orally 2 times a day for 14 days. and the control group were administered a placebo. The mandibular condyle heads were sectioned in 5 §­ thickness. The specimens were stained with H-E staining. COX-2 immunohistochemical staining and IGF-¥° immunohistochemical staining and examined under light microscope. After TRAP staining. the number of chondroclasts were calculated. The observed results were as follows: Indomethacin inhibited expression and distribution of COX-2 and IGF-¥° on the proliferative zone of condylar cartilage. Indomethacin decreased the number of chondroclastes on the mineralization zone by a time-dependent manner (p<0.05). Indomethacin inhibited expression and distribution of IGF-¥° by a dose and time-dependent manner. These results show that indomethacin inhibited expression and distribution of COX-2 and IGF-¥° on the proliferative zone of condylar cartilage and decreased the number of chondroclasts and suggests that when indomethacin is administered for a long time. condyle growth could be delayed.
